Understanding the processes occurring during receptor activation is of fundamental importance to pharmacology, and activation of receptors such as the NMDA receptor, which have an integral membrane ion channel, has proved to be particularly amenable to study using single channel recording techniques. The activation of muscle and Torpedo acetylcholine receptors (AChRs) has been studied in great detail. Using these approaches the rates of the reactions involved in receptor activation have been estimated and for the endplate AChRs these rates are consistent with the observed rate of synaptic transmission at the neuromuscular junction. This has led to the expectation that new insights into the processes underlying the NMDA receptor-mediated component of excitatory post-synaptic currents (NMDA EPSCs) will be obtained from studies of NMDA receptor single channels.
