A sensitive method (single cell gel (SCG) electrophoresis) to examine DNA damage and repair at individual cell level. Single cells are embedded in agarose on a microscope slide, are lysed and then electrophoresed in situ. The slide is then stained with fluorescent dye (e.g. acridine orange) and examined under the microscope. Intact DNA remains in the cell, as the comet's ‘head’, fragments of different sizes form the tail.
