A protein (NACHT, LRR, and PYD domains-containing protein 3, NALP3, cold autoinflammatory syndrome 1 protein, PYRIN-containing APAF1-like protein-1, angiotensin/vasopressin receptor AII/AVP-like, 1034 aa) that is strongly expressed in polymorphonuclear cells and osteoblasts. It interacts selectively with ASC, may be an upstream activator of NFκB signalling, and activates caspase-1 in response to a number of triggers including bacterial or viral infection which leads to processing and release of IL-1B and IL-18. Mutations cause familial cold urticaria (familial cold autoinflammatory syndrome type 1), Muckle–Wells syndrome, and chronic infantile neurological cutaneous and articular syndrome.
