4′,6-diamidino-2-phenylindole, a fluorescent dye that binds to DNA. DAPI-staining of chromosomes within nuclei can be followed with the collection of three dimensional data sets obtained by recording serial images at 0.25 μm intervals. From these, linearized maps of all the chromosomes can be constructed. The structure of the DAPI molecule is shown below.
