The major component of teeth, a structural biocomposite of needle-like crystals of hydroxyapatite embedded in a fibrous collagen matrix, very similar to compact bone but secreted by odontoblasts. Dentin matrix protein (DMP-1, 513 aa) is a serine-rich acidic matrix protein that has a key role in mineralization. Oligomers of DMP-1 temporarily stabilize calcium phosphate nanoparticle precursors by sequestering them and preventing their further aggregation and precipitation. It binds to RGD sequence-dependent integrins. Dentin sialophosphoprotein (DSPP, 1301 aa) is cleaved to produce two major acidic matrix proteins of dentin, dentin sialoprotein (DSP, 447 aa) and dentin phosphoprotein (DPP, phosphophoryn, 839 aa). Mutations in DSPP are a causative factor in dentinogenesis imperfecta.
