The spontaneous loss of purine residues from DNA, which occurs at an appreciable rate in vivo (estimated to be ~10 000 per cell per day). Although depurination does not disrupt the structural integrity of the phosphodiester backbone of DNA, constant repair by the base excision repair pathways is required to avoid irreversible loss of information.
