The proposition that cell proliferation in culture is rate-limited by the diffusion of some essential factor, probably a growth factor, from the bulk medium and through a diffusion boundary layer (boundary layer hypothesis) immediately adjacent to the cell surface. By spreading the cell increases its capacity to obtain sufficient of this critical factor and once cells become crowded they will not progress from G0 to G1 in the cell cycle—a density dependent inhibition of proliferation.
