In principle, the polymerization of deoxyribonucleotide triphosphates to form DNA which in vivo occurs mostly as template-directed DNA replication although some repair synthesis does occur. In retroviruses, DNA synthesis is directed by an RNA template and the reverse transcriptase enzyme has been used in vitro to produce DNA (complementary DNA; cDNA) from mRNA. In vitro synthesis can also be done by sequential operator-controlled reactions that generate an oligonucleotide of the desired sequence.
