One of the family of giant proteins that associate with the F-actin cytoskeleton and with the nuclear envelope. The human enaptin gene spreads over 515 kb and gives rise to several splicing isoforms (nesprin-1, myne-1, synaptic nuclear envelope protein 1 (syne-1), CPG2), the largest of which is predicted to be >1 MDa. It is highly homologous to nuance (nucleus and actin connecting element, SYNE2, nesprin-2) and has multiple spectrin repeats, a nuclear localization signal, a transmembrane domain within the C-terminal Klarsicht homology region, and several N-glycosylation and phosphorylation sites.
