A family of ten G-protein-coupled receptors involved in the wnt signalling pathway and other signalling pathways. Ligand binding induces activation of dishevelled proteins in the cytoplasm and eventually to activation of transcription factors. Variation in the cytoplasmic domain links the receptors to various pathways. Mutations in frizzled 4 (FRZ4, 537 aa) lead to exudative vitreoretinopathy; the FZD9 gene is located within the Williams–Beuren syndrome common deletion region. Secreted frizzled-related proteins (SFRPs) appear to modulate Wnt signalling by competing with membrane-bound frizzled receptors for the binding of secreted Wnt ligands.
