Oncogenes (glioma-associated oncogenes) that encode zinc finger transcription factors of the Krüppel family. GLI1 (1106 aa) is colocalized in cytoplasm with SUFU. In mice three related proteins, Gli1, Gli2, and Gli3, have been implicated in the transduction of sonic hedgehog signalling. A variety of disorders are associated with mutations in the Gli transcription factors, including foregut malformations. Mutations in gli2 are associated with basal cell carcinomata and holoprosencephaly, mutations in gli3 are found in Pallister–Hall syndrome and Greig cephalopolysyndactyly syndrome.
