Genes that encode transmembrane proteins of the endoplasmic reticulum (insig-1, 277 aa; insig-2, 225 aa) that bind the sterol-sensing domain of the SREBP cleavage-activating protein (SCAP) and retain the SREPB–SCAP complex in the ER. This prevents proteolytic cleavage of SREBP to generate the active nuclear form. They are thought to play a central role in cholesterol homeostasis.
