A multidomain guanine nucleotide exchange factor (P-CIP10, huntingtin-associated protein-interacting protein, DUO), one of the Dbl family, that interacts with small GTP-binding proteins of the rho family. Multiple kalirin isoforms with different functional domains are expressed in the brain and the gene has 60 coding exons. Kalirin-7 localizes to the postsynaptic density and is involved in formation of dendritic spines. Other isoforms associate with secretory granules and through the interaction with G proteins are involved in regulating actin organization, endocytosis, exocytosis, and free-radical production. Single-nucleotide polymorphisms in the first intron of an alternative transcript of the gene are associated with susceptibility to early-onset coronary artery disease.
