A method analogous to the polymerase chain reaction (PCR) used to detect small quantities of a target DNA. The method relies upon the requirement, by DNA ligase, for sequence similarity to allow ligation of synthetic oligonucleotides (probe) bound to target DNA. Various ligase-based techniques have been developed for the high-throughput detection of DNA, homogeneous assays, single-molecular detection, and scanning of unknown mutations.
