A graphical method for deriving Vmax (maximal reaction rate) and Km (rate constant, Michaelis–Menten constant) for an enzyme-catalysed reaction. The reciprocal of the reaction rate (1/V) is plotted against the reciprocal of the substrate concentration (1/S) and the y-intercept is equivalent to the inverse of Vmax and the x-intercept represents −1/Km. Because the method gives undue weight to the least accurate points, other methods of analysis, such as the Eadie–Hofstee plot, are preferable.
