A family of MAGUK proteins (membrane-associated guanylate kinase inverted proteins) with an inverted arrangement of protein–protein interaction domains. MAGI-1 is present at adherens and tight junctions, where it acts as a structural and signalling scaffold and associates with megalin and synaptopodin. MAGI-2 (atrophin-1-interacting protein-1, AIP1) coimmunoprecipitates with stargazin and interacts with PTEN. Mutations may be responsible for infantile spasms in some Williams–Beuren syndrome patients. MAGI-3 has been found in mouse and interacts with the TGFα cytoplasmic region.
