A family of three cotranscriptional regulators (mastermind-like proteins) that bind to the ankyrin repeat domain of Notch and are essential for signalling. They have distinct tissue-specific distributions; MAML1 (1016 aa) localizes to nuclear bodies and interacts with MEF2C. MAML2 (1173 aa) is widely expressed. A MAML2/*MECT1 fusion gene arising from a chromosomal translocation characterizes mucoepidermoid carcinoma (a common malignant salivary gland tumour). MAML3 (1133 aa) acts selectively on different notch promoters.
