A family of serine/threonine kinases (mixed lineage kinases, MAP3K9, 10, 11) that regulate signalling by the JNK and p38 MAPK pathways. They are considered to be mitogen-activated protein kinase kinase kinases (MAPKKKs, MAP3Ks). There are two subgroups, one containing the highly related MLK1 (MAP3K9), MLK2/MST (MAP3K10), and MLK3/SPRK/PTK (MAP3K11) kinases. Overexpression of MLKs effectively induces apoptotic death of cultured neuronal PC12 cells
