Signalling molecules involved in the development of innate immunity against intracellular pathogens through activation of inflammatory mediator pathways. The adaptor protein ASC links pathogen recognition by pyrin domain (PYD)-containing proteins (PYD-NLR, PAN, PYPAF, NALP, Nod, and Caterpillar proteins), to the activation of downstream effectors. Activation of these effectors occurs when specific protein complexes, known as inflammasomes, are formed. The pyrin domain is involved in protein–protein interactions and is a subfamily of the death domain (DD) superfamily.
