A cysteine endopeptidase (separase, Esp1, EC 3.4.22.49, 2120 aa), related to caspases, that cleaves the RAD21 component of the cohesin complex that holds sister chromatids together, allowing the chromatids to separate in anaphase. It is inhibited by securin and this inhibition is relieved when the anaphase-promoting complex ubiquitinates securin leading to its degradation.
