A technique in which the genomic DNA of an organism is randomly broken into smaller fragments by mechanical shearing or partial digestion with restriction endonucleases (q.v.) and cloned in appropriate vectors to produce a genomic library (q.v.). The clones are then used in shotgun sequencing (q.v.).
