The post-transcriptional modification of a protein by the addition of small ubiquitin-related modifier (sumo) proteins. The effect may be to stabilize or to alter subcellular localization. SUMO-1 (101 aa) conjugates as a monomer, SUMO-2 (95 aa) and SUMO-3 (103 aa), which are closely related, are conjugated as higher molecular weight polymers. Targets include p53, ran-Gap. Several transcription factors (e.g. C/EBP and c-myb) are regulated by sumoylation.
