A non-receptor tyrosine kinase (EC 2.7.1.112, 1187 aa) upstream of STAT3, important in the initiation of type I interferon signalling and other cytokine signalling pathways and an important regulator of lymphoid tumour surveillance. Deficiency of tyk2 causes autosomal recessive hyper-IgE syndrome (HIES) with atypical mycobacteriosis, a primary immunodeficiency characterized by recurrent skin abscesses, pneumonia, and elevated serum IgE.
