A family of guanine nucleotide exchange factors (GEFs) (vav1, vav2, vav3) for the rho/rac GTPases, members of the dbl family. The vav1 proto-oncogene product (p95vav, 845 aa) is found mainly in haematopoietic cells but vav2 (872 aa) has a wider tissue distribution. Activation of vav1 by phosphorylation in Bcr-Abl-expressing cells leads to the activation of Rac-1 and is associated with leukaemias. Vav3 (847 aa) is expressed at higher levels in glioblastoma vs low-grade glioma and is overexpressed in prostate cancer.
