As librarian of Cambridge University Library from 1629 to 1653, Wheelocke increased its holdings twelvefold. In 1632, Wheelocke became England's first professor of Arabic; in 1640, he became professor of Anglo-Saxon, the study of which he advanced by his publication of the first editions of the Old English translations of Bede's Historia Ecclesiastica and of the Anglo-Saxon Chronicle (1643).
